Bacteries associees aux production de Brachionus plicatilis ArchiMer

Bacteria associated with semi-continuous culture of Brachionus plicatilis , which is used as the first food of marine fish larvae, were counted on Zobell 2216 E medium and identified. The numbers of bacteria were very high in both Brachionus (about 10 super(11) bact. per ml of digestive content) and in water (from 1.6.10 super(6) to 4.7.10 super(7) bact. per ml). The cultures were studied at driving a time when mortalities occurred frequently. The introduction of a Pseudomonas) phenon at a proportion of about 50% to the dominant microflora persisted in time and was little influenced by diet. Comparisons between continuous and batch processing to produce clavulanic acid by Streptomyces clavuligerus BABT

The aim of the present work was to compare CA production in continuous culture with and without cell recycling and in batch process by Streptomyces clavuligerus. Continuous cultivations with high cell concentration using cell recycling were performed utilizing a hollow fiber ultrafiltration module to separate cells from the filtrate broth. The continuous cultures without cell recycling and the batch cultivations were performed conventionally. The highest productivity was attained in the continuous cultivation with cell recycling (22.2 mg.L-1.h-1). The highest CA concentration was obtained in the batch process (470 mg.L-1.h-1).

Cultivation of an immobilized (hyper)thermophilic marine microbial community in a bioreactor ArchiMer

Cultivation in a bioreactor of immobilized deep-sea hydrothermal microbial community was tested in order to assess the stability and reactivity of this new system. A community composed of 8 hydrothermal strains was entrapped in a polymer matrix that was used to inoculate a continuous culture in a gas-lift bioreactor. The continuous culture was performed for 41 days at successively 60°C, 55°C, 60°C, 85°C and 60°C, at pH 6.5, in anaerobic condition and constant dilution rate. Oxic stress and pH variations were tested at the beginning of the incubation. Despite these detrimental conditions, 3 strains including 2 strict anaerobes were maintained in the bioreactor. Etude de l’influence de la concentration et de la spéciation de trace de cadmium sur la croissance et la physiologie d’une espèce phytoplanctonique ArchiMer

The aim of this study is to establish a continuous culture of a diatom (Phaeodactylum tricornutum) in a photobioreactor. As soon as the culture is stable and regulated, increasing cadmium amounts are added to the culture to induce a metal stress in order to study its influence on the production of Volatile Reduced Sulphur Compounds (H2S, OCS, MeSH, DMS and DMDS), and on the production of ligands. In this report, the methods used to assess the continuous culture and to analyse RVSC and other ligands production are described. J This study allowed us to assess cadmium impact on the VRSC and on the production of ligands.

Experimental systems for the study of bacterial degradation of pollutants from the oil industry. ArchiMer

The release of pollutants into the environment, either during normal operations or due to accidental discharges, has focussed attention on the fate and effects of xenobiotic compounds in the marine environment. This interest has been heightened by the development of the off-shore industry with the subsequent discharge of biocides from platforms. This paper describes the results obtained from investigations into the fate of biocides in the marine environment using continuous-culture techniques.

Since the discovery of hydrothermal vents, multiple studies have been conducted in order to study microbial diversity. Molecular inventories realized have thus demonstrated a great diversity of species that contrasts with the low proportion (1%) of species isolated by culture approach. A new cell immobilization approach by inclusion in a polymer matrix (gellan and xanthan) has been developed for the study of these thermophilic anaerobic marine communities. The system, based on the formation of an emulsion between an inoculated polymer solution and oil, allows the entrapment of cells in gel beads with a diameter between 1 and 2 mm. An unstructured model for growth and lactic acid production during two-stage continuous cultures of Lactobacillus helveticus was previously developed. The Verlhust model was considered to describe growth kinetics. Physiology and continuous culture of the hyperthermophilic deep-sea vent archaeon Pyrococcus abyssi ST549 ArchiMer

The deep-sea vent archaeon Pyrococcus abyssi strain ST549 was grown in batch cultures in closed bottles and by continuous culture in a gas-lift bioreactor, both in the presence and in the absence of elemental sulfur. Growth on carbohydrates, proteinaceous substrates and amino acids was investigated. The disaccharides maltose and cellobiose were shown not to be able to enhance growth suggesting that P. abyssi ST549 is unable to use them as carbon sources. By contrast, proteinaceous substrates such as peptone and brain heart infusion were shown to be very good substrates for the growth of P. abyssi ST549 and allowed growth at high steady-state cell densities in continuous culture. Stimulation of the Megasphaera elsdenii’s butyrate production in continuous culture by a yeast additive BABT

Continuous culture study of the ruminal bacterium Megasphaera elsdenii using lactate as carbon source is reported. Butyrate, the minor product in batch culture, was the main product in continuous culture. Under steady state conditions the biomass concentration was increased from 0.46 to 0.72 g l-1. Addition of Yea Sacc did not result an increased availability of carbon. The produced amounts of valerate and CO2 were calculated by using a combined carbon and degree of reduction balances. According to these results, the lactate catabolism of M. elsdenii was changed by the presence of yeast additive, yielding more butyrate and diminishing the production of valerate, while acetate and propionate production was not affected. Transient initial phase in continuous culture of Isochrysis galbana affinis Tahiti ArchiMer

Starting microalgae continuous culture generally includes a preliminary batch culture to obtain sufficient cell density. It is possible to apply continuous regime from the beginning of the culture (IC mode) rather than to begin by an initial batch (IB mode). It is our purpose to check that both initial modes lead to identical steady-state cell characteristics. The microalga Isochrysis galbana affinis Tahiti was used for this comparative study. With an initial cell density of 5 x 10(5) cell ml(-1) and a dilution rate of about 1.0 d(-1), both IB and IC modes led to identical cell density once steady-state is reached between 6 and 8 d after inoculation in the two cases.
